Nobilis SP Antigen

This antigen is used for the detection of antibiodies due to infections caused by both standard and variant strains of Salmonella pullorum and Salmonella gallinarum in blood or serum of chickens and turkeys by the plate agglutination test.  
When blood or serum is mixed with the test antigen, agglutination occurs in the presence of specific antibodies. The staining of the test antigen renders this process more easily visible.

The test should be used as a flock test. Antibodies due to infections caused by other Salmonella with 0-antigens, equal to those of Salmonella pullorum (like Salmonella enteritidis) may be detected by the test as well. As the antigen is not used in vivo, there is no requirement for a with-holding period, thus:

Materials:

• A well cleaned glassplate, illuminated from below (avoid excessive heat from the light source).
• A small stirring rod.

The test should be carried out between 20°C and 25°C. A drop of serum of approximately equal size is added to a drop of the well-shaken antigen. The drops are then mixed with a stirring rod and spread to a circular area of -1,5cm diameter. By rocking the plate a positive agglutination is stimulated. The reaction should be carried out with fresh serum. The serum should not be frozen as this may lead to aspecific reactions.

Test interpretation
A positive reaction is indicated by agglutination within 2 minutes after mixing.
This agglutination reaction may be delayed at temperatures below +2°C.

Presentation
10 ml vials (± 200 tests).

Storage
Store between 2-8°C. Do not freeze.Protect from light. Discard unused antigen after vial has been opened.

Further information
Do not use coagulated blood.
The stirring rod should be rinsed after each sample.

Disposal:
This is a biological product which requires disinfection after use. After completion of the test the agglutination plates should be soaked for 5 minutes in 70% alcohol or 750 ppm iodophor, then washed in water. Opened vials should be disinfected by adding disinfectant to the contents and then treating as above. Washing water should be disposed of to a closed drain and flushed away with copious quantities of water.
